Profile of Yigong Shi.

چکیده

Growing up in rural China under the imposed hardships of the cultural revolution, Yigong Shi did not have the luxury of toys. Instead Shi, now the dean of the School of Life Sciences at Tsinghua University in Beijing, made elaborate models out of clay he collected from the fields. “I would stay for hours to a make a ship or train model with small parts,” he recalls. Decades later, as a structural biologist, Shi is still making models, albeit with different tools. Elected to the National Academy of Sciences in 2013, Shi studies the molecular mechanisms that govern the awakening of apoptotic enzymes in worms, fruit flies, and mammals, seeking to illuminate function by revealing structure. Apoptosis, or programmed cell death, is a process akin to cellular containment and cleaning. An evolutionarily conserved cascade of enzyme activity triggers this tightly orchestrated cellular dismantling, which is especially important during embryonic development and tumor formation. In his Inaugural Article (1), Shi examines the fundamental mechanism of the final enzyme in the cascade, offering what he calls “a somewhat controversial conclusion” regarding caspase activation. He demonstrated that the enzyme caspase-9, which controls cell death triggered by intracellular stress signals, is awakened in a complex process that requires the cooperation of another protein, Apaf-1.

Allosteric Activation of a Bacterial Stress Sensor

In Gram-negative bacteria, envelope stress signals such as unfolded outer membrane proteins (OMP) activate the periplasmic protease DegS. This protease then triggers a cellular pathway to alleviate the stress. Now Sohn et al. (2007) show conclusively that inhibition of DegS is relieved allosterically by binding of the C-terminal sequences in unfolded OMPs to the PDZ domain of DegS.

China's research culture.

Government research funds in China have been growing at an annual rate of more than 20%, exceeding even the expectations of China's most enthusiastic scientists. In theory, this could allow China to make truly outstanding progress in science and research, complementing the nation's economic success. In reality, however, rampant problems in research funding—some attributable to the system and ot...

Caspase Activation Revisiting the Induced Proximity Model

Caspases execute cell death. The mechanism of effector caspase activation primarily involves reorganization of active site loops following the activation cleavage. The Induced Proximity hypothesis, originally proposed to explain the activation of initiator caspases, has recently been reinterpreted to be proximity-driven dimerization of initiator caspases, and consequently their activation. The ...

Do public work schemes deter or encourage outmigration? Empirical evidence from China

How does the introduction of rural public work schemes impact individual incentives to migrate? This paper examines this question in the context of rural public work program (Yigong-daizhen) in China, and unveils empirical evidence that suggest that the introduction of Yigong-daizhen projects in fact stimulates outmigration at the village level, after controlling for village characteristics, pr...
